近日,一項(xiàng)發(fā)表于雜志The Journal of Cell BioLogy上的研究論文中,來自加尼弗尼亞大學(xué)的研究人員通過研究發(fā)現(xiàn),一種幫助維持胚胎干細(xì)胞(ESCs)身份的特殊蛋白或可促進(jìn)干細(xì)胞的DNA修復(fù),研究者表示,這種名為Sall4的蛋白質(zhì)在癌細(xì)胞中也扮演著類似的角色,其可以幫助修復(fù)癌細(xì)胞的DNA損傷從而使得癌細(xì)胞免于化療殺傷。
修復(fù)破碎的DNA對(duì)于ESCs來說非常重要,因?yàn)楦杉?xì)胞會(huì)將任何突變傳遞給分化形成的細(xì)胞,而小鼠的ESCs就非常善于進(jìn)行DNA的修復(fù),相比其它分化細(xì)胞而言小鼠的ESCs攜帶的突變會(huì)相對(duì)較少,但其進(jìn)行DNA損傷修復(fù)的機(jī)制目前尚不清楚;于是研究人員就開始檢測(cè)是否一直ESCs分化的蛋白Sall4在DNA修復(fù)過程中扮演著重要的角色。
研究者表示,缺失Sall4的ESCs并不擅長于修復(fù)DNA的雙鏈破碎,而雙鏈破碎是DNA損傷的一種災(zāi)難性的破壞形式。研究者同時(shí)還觀察到,當(dāng)誘導(dǎo)小鼠ESCs發(fā)生DNA損傷后,和Sall4相關(guān)的蛋白質(zhì)就會(huì)參與進(jìn)行DNA的修復(fù)。
In a recent research paper published in the journal The Cell BioLogy, researchers from the University of California discovered through research that a special protein that helps maintain the identity of embryonic stem cells (ESCs) or promotes stem cells DNA repair, the researchers said, the protein called Sall4 also plays a similar role in cancer cells, which can help repair DNA damage in cancer cells so that cancer cells from chemotherapy kill.
Repairing broken DNA is important for ESCs as stem cells pass any mutation to differentiated cells, and mice 'ESCs are very good at DNA repair, as are the other ESCs that mice carry The mutation will be relatively small, but its mechanism for DNA damage repair is not clear; so the researchers began to test whether the ESCs have been differentiated protein Sall4 plays an important role in DNA repair process.
The researchers said ESCs lacking Sall4 are not good at repairing double-stranded DNA, a disastrous form of DNA damage. The researchers also observed that, when DNA damage was induced in mouse ESCs, proteins associated with Sall4 were involved in DNA repair.
