生物活性:Cytochalasin D (Zygosporin A) 是一種有效的肌動(dòng)蛋白聚合抑制劑,可能來(lái)源于真菌。細(xì)胞松弛素 D 具有細(xì)胞滲透活性。細(xì)胞松弛素 D 通過(guò)與 G-肌動(dòng)蛋白結(jié)合來(lái)抑制 G-肌動(dòng)蛋白-肌動(dòng)蛋白絲切蛋白相互作用。 Cytochalasin D 還抑制 cofilin 與 F-肌動(dòng)蛋白的結(jié)合,并降低活細(xì)胞中肌動(dòng)蛋白聚合和解聚的速率。細(xì)胞松弛素 D 可以減少外泌體的釋放,進(jìn)而減少腫瘤環(huán)境中存在的存活蛋白的數(shù)量。細(xì)胞松弛素 D 誘導(dǎo) Yap[1][2][3][4] 的磷酸化和細(xì)胞質(zhì)滯留。
體外:Cytochalasin D(3 和 10 μM;30 分鐘)導(dǎo)致 COS-7 細(xì)胞收縮和分枝,以及 F-肌動(dòng)蛋白從長(zhǎng)纖維轉(zhuǎn)化為點(diǎn)狀結(jié)構(gòu)[1]。
Cytochalasin D( 0.3、1、3 和 10 μM;30 分鐘)濃度依賴性地降低 COS-7 細(xì)胞中肌動(dòng)蛋白解聚的速率[1]。
細(xì)胞松弛素 D(1 μM;NIH3T3 細(xì)胞)破壞應(yīng)力纖維,誘導(dǎo) Yap 磷酸化,同時(shí)細(xì)胞保持原有區(qū)域并丟失核 Yap 定位[4]。
