巰基反應(yīng)性熒光染料Cy5.5馬來酰亞胺
Cyanine 5.5應(yīng)用非常廣泛,不僅可用于生物大分子的標(biāo)記,還被用作一些小分子的衍生化試劑。maleimide(馬來酰亞胺)常被用來標(biāo)記巰基,Cyanine5.5 Maleimide與其他標(biāo)記巰基的熒光試劑(如FITC,TRITC等)相比,它反應(yīng)選擇更高。Cyanine5.5 Maleimide是近紅外染料,除了用于標(biāo)記多肽,蛋白和寡核苷酸氨基外,也常被用于小動物活體成像。因?yàn)樗荛_了很多物質(zhì)都有吸收的可見光區(qū),吸收的近紅外光在生物組織中的穿透深度較大,而激發(fā)的熒光受生物組織本底的影響較少,所以能夠在深層組織產(chǎn)生信號。尤其是在700~900 nm的范圍中,水和血紅蛋白的吸收都很少,近紅外光可以深入組織內(nèi)部多達(dá)15 cm。同時,這類染料還擁有紫外光區(qū)染料和同位素標(biāo)記無法具備的生物安全性。Cyanine 5.5 (cy 5.5)是常用的近紅外熒光染料。
cyanine 5.5 Maleimide需要共溶劑,但由于其疏水性比Cyanine7弱,應(yīng)用更為廣泛。陜西新研博美生物提供三乙胺鹽酸鹽(triethylammonium salts)形式的cyanine 5.5 Maleimide,較其他公司鉀鹽形式的cyanine 5.5 Maleimide更易容,在光學(xué)性質(zhì),反應(yīng)性相同的情況下,擁有更好的溶解性。
cyanine 5.5 Maleimide(馬來酰亞胺)廣譜學(xué)性質(zhì)與Alexa Fluor 680 Maleimide和DyLight 680 Maleimide類似,可作為其替代物。
