【預(yù)期用途】
流感病毒可分為甲(A)、乙(B)、丙(C)三型。其中,甲型流感依據(jù)流感病毒血凝素蛋白(HA)的不同可分為1-16種亞型,根據(jù)病毒神經(jīng)氨酸酶蛋白(NA)的不同可分為1-9種亞型,HA不同亞型可以與NA的不同亞型相互組合形成不同的流感病毒。而qin類特別是水qin是所有這些流感病毒的自然宿主,H7N9qin流感病毒是其中的一種。H7N9流感病毒既可以感染qin類,也可以感染人。人感染H7N9qin流感會(huì)出現(xiàn)嚴(yán)重肺炎,癥狀包括發(fā)燒、咳嗽、呼吸困難等。
本試劑盒適用于檢測(cè)呼吸道樣本、血清、肺組織等樣本中H7N9qin流感病毒RNA,可用于該病毒感染的實(shí)驗(yàn)室診斷和宿主動(dòng)物的監(jiān)控。檢測(cè)結(jié)果僅供研究,不用于臨床診斷。
【檢驗(yàn)原理】
本試劑盒基于實(shí)時(shí)熒光PCR技術(shù),分別選取H7N9qin流感病毒的HA基因和NA基因保守區(qū)作為擴(kuò)增靶區(qū)域,設(shè)計(jì)特異性引物探針,通過一步法實(shí)時(shí)熒光PCR體系擴(kuò)增對(duì)H7N9qin流感病毒進(jìn)行定性檢測(cè)。反應(yīng)體系中除特異性引物和特異性熒光探針外,還配以對(duì)應(yīng)的PCR反應(yīng)Buffer、逆轉(zhuǎn)錄酶、Hot-Start Taq酶、核苷酸單體(dNTPs)、Mg2+等成分,可實(shí)現(xiàn)對(duì)H7N9qin流感病毒核酸靈敏特異地檢測(cè)。
