現(xiàn)代色譜分離對色譜柱提出更高的要求,如:廣泛的通用性,對酸性,堿性和中性的樣品都能對稱出峰,填料能夠承受更寬的PH使用范圍,可以方便的選擇色譜分離的*條件。填料具備更高的機(jī)械強(qiáng)度,保證色譜柱更長的壽命。而決定液相色譜柱性能的關(guān)鍵在于使用硅膠的品質(zhì)。傳統(tǒng)的液相色譜柱所填裝的硅膠,由于硅球表面的缺陷,或是其內(nèi)部的金屬雜質(zhì)含量較高,使下一步的化學(xué)鍵合不能*進(jìn)行。因此會在硅球的表面殘余一定的硅醇基和其他活性基團(tuán),直接造成色譜柱在分析樣品,特別是堿性樣品時,出現(xiàn)脫尾現(xiàn)象或樣品被吸附。 |
