NUCLEOSIL Chiral-1配位交換手性柱
配位交換色譜LEC(ligand exchange chromatography)的分離原理是,被測手性樣品與色譜柱的官能團中的金屬陽離子(CU2+)產(chǎn)生三重混合配位交換,而L-羥基-脯氨酸產(chǎn)生的不對稱中心導(dǎo)致了對樣品的手性色譜拆分。NUCLEOSIL CHIRAL-1 配位交換手性柱色譜條件的選擇我們建議使用CUSO4的水溶液作為NUCLEOSIL CHIRAL-1的流動相,并可以按一下原則通過改變流動相條件改善分離效果。
Chiral-2/3刷型交換手性柱
NUCLEOSIL CHIRAL-2/3手性柱是正相原理分離手性化合物的。它是在Nucleosil硅球上交聯(lián)鍵合N-(3,5-二硝基苯酰)-“D,L”-苯基甘氨酸作為手性官能團,分離的原理是刷型交換(brush type)。這種分離原理主要用來控制農(nóng)藥或藥物的生產(chǎn)工藝中間體的光學(xué)純度。
由于NUCLEOSIL CHIRAL-2使用的官能團是D-苯基甘氨酸,而NUCLEOSIL CHIRAL-3使用的官能團是L-苯基甘氨酸,使兩種手性柱在分離同一手性化合物時,出峰順序正好相反。這在質(zhì)檢控制產(chǎn)品光學(xué)純度時是十分重要的,我們可以選擇NUCLEOSIL CHIRAL-2或CHIRAL-3,讓雜質(zhì)在主要成份前出峰,避免由于脫尾造成定量的不準確。色譜柱使用條件:
NUCLEOSIL CHIRAL-2/3手性柱使用的流動相一般是非極性有機溶劑(正乙烷,異辛烷),添加極性有機溶劑(四氫呋喃,醇類,氯代烴)和極少量強酸(三fuyisuan)。強酸通常能夠明顯的改善手性樣品的分離。而被測的堿性樣品,需要通過苯甲酰氯或3,5-二硝基苯甲酰氯衍生化后進行檢測。
